Anti-IL12B scFv-Fc is a recombinant antibody targeting the p40 subunit (IL12B) of both Interleukin-12 (IL-12) and Interleukin-23 (IL-23), key cytokines involved in Th1 and Th17 immune responses. The scFv provides specific binding to IL12B, while the Fc region enhances stability and potential for dimerization. This format is being developed to block the activity of both IL-12 and IL-23, aiming to modulate inflammation in autoimmune and inflammatory diseases.

- The potency of the purifed monoclonal antibody in inhibiting interleukin-12 (IL-12) activity was assessed using the phytohaemagglutinin (PHA) blast assay. The half-maximal inhibit concentration (IC₅₀) values derived from this assay indicated that human IL-12 was inhibited at an IC₅₀ of approximately 0.31 nM, while the corresponding value for cynomolgus monkey IL-12 was approximately 1.4 nM.
The optimal dilution concentration is determined by the experiment.

Associates with IL23A to form the IL-23 interleukin, a heterodimeric cytokine which functions in innate and adaptive immunity.
IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues.
IL-23 binds to a heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of pro-inflammatory cytokines.
IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis.
