The Rat Pecam1 Quick ELISA Kit (VS-0726-YC921) is designed for quickly quantify Rat Platelet endothelial cell adhesion molecule with a sensitivity of 0.938 ng/mL. The kit is based on sandwich ELISA and the total experimental duration is 120 minutes. It is a reliable tool used for the quantitative detection of Pecam1 in Serum, Plasma, Cell culture supernatant, Cell lysate, Tissue lysate and other biological samples. PECAM1 (CD31) localizes to endothelial cell-cell junctions, mediating homophilic interactions required for leukocyte transendothelial migration and vascular integrity maintenance. It serves as the standard clinical immunohistochemical biomarker for quantifying tumor angiogenesis.

- Assay Principle
- To perform the assay, secure the required microplate wells and dispense 50 μL of the antibody cocktail (capture/detection antibody) into each well, followed immediately by 50 μL of Standard or Sample. To prevent cross-contamination, use fresh pipette tips for each sample and standard, touching the tip lightly to the liquid level during dispensing. Mix thoroughly by gently tapping the plate for 10 seconds, then seal and incubate statically for 60 minutes at 37°C. Following incubation, aspirate the wells and wash the plate twice, ensuring the liquid is decanted completely after each cycle without soaking. Next, add 100 μL of HRP-Streptavidin Conjugate to each well, seal the plate, and incubate statically for 30 minutes at 37°C. Repeat the wash cycle, washing the plate 5 times without soaking. Subsequently, add 90 μL of TMB Substrate Solution, seal the plate, and incubate statically at 37°C for 10 to 20 minutes, monitoring color development closely to prevent overdevelopment. Finally, halt the reaction by adding 50 μL of Stop Solution to each well, and immediately measure the optical density (OD) at 450 nm to calculate the analyte concentration.
