Mycoplasmas (1x105 CFU/ml) were incubated with PBS (positive control), boiled CA27 (negative control, 3 μg/ml), or active CA27 (1–3 μg/ml), added to Huh7 cells, and then cultured for 2 days. Mycoplasma infection of Huh7 cells was then measured by flow cytometric analysis with CA27.

This research investigates how Mycoplasma hyorhinis infection influences esophageal squamous cell carcinoma (ESCC) progression and develops a novel therapeutic approach using DNA aptamers. The study demonstrates that M. hyorhinis infection promotes the malignancy of ESCC cells by enhancing migration, invasion, and metastasis capabilities. Using cell-SELEX technology, researchers generated a single-stranded DNA aptamer called ZY3A that specifically targets the M. hyorhinis-encoded p37 protein. The aptamer works by blocking the interaction between p37 protein and its receptor TLR4 on ESCC cell membranes, effectively neutralizing M. hyorhinis infection and inhibiting cancer cell migration, invasion, and metastasis both in vitro and in vivo.
Creative Biolabs provided a monoclonal anti-mycoplasma p37 protein antibody (HPAB-0208-YJ) that was crucial for identifying and confirming the binding target of the aptamer ZY3A. This antibody enabled researchers to validate that p37 was the specific target protein and helped elucidate the mechanism through which the aptamer neutralized M. hyorhinis infection. The antibody's specificity was essential for the pulldown assays, western blot analysis, and immunofluorescence studies that demonstrated how ZY3A blocks the interaction between p37 and TLR4, providing key evidence for developing ZY3A as a potential therapeutic tool against M. hyorhinis-associated ESCC.
