ACO2: A Key Enzyme in Neurodegeneration Metabolic Dysfunction and Cancer
ACO2 encodes a critical mitochondrial enzyme involved in the tricarboxylic acid (TCA) cycle, and its dysfunction has been linked to a range of neurological and metabolic diseases. Deficiency in its activity can impair cellular energy production, particularly in energy-demanding tissues such as the brain and muscles. Mutations have been associated with progressive neurodegenerative disorders marked by symptoms like optic atrophy, ataxia, and cognitive decline, likely due to compromised mitochondrial function and increased oxidative stress. In inherited metabolic syndromes, reduced enzyme efficiency can lead to lactate accumulation and disturbed energy homeostasis. Emerging research also suggests a potential link to tumor metabolism, as altered mitochondrial pathways support the growth and survival of cancer cells under metabolic stress. In the context of age-related diseases, decreased expression has been implicated in mitochondrial dysfunction contributing to conditions such as Alzheimer's and Parkinson's. Background
The protein encoded by this gene belongs to the aconitase/IPM isomerase family. It is an enzyme that catalyzes the interconversion of citrate to isocitrate via cis-aconitate in the second step of the TCA cycle. This protein is encoded in the nucleus and functions in the mitochondrion. It was found to be one of the mitochondrial matrix proteins that are preferentially degraded by the serine protease 15(PRSS15), also known as Lon protease, after oxidative modification.
