ACO2: A Key Enzyme in Neurodegeneration Metabolic Dysfunction and Cancer

ACO2 encodes a critical mitochondrial enzyme involved in the tricarboxylic acid (TCA) cycle, and its dysfunction has been linked to a range of neurological and metabolic diseases. Deficiency in its activity can impair cellular energy production, particularly in energy-demanding tissues such as the brain and muscles. Mutations have been associated with progressive neurodegenerative disorders marked by symptoms like optic atrophy, ataxia, and cognitive decline, likely due to compromised mitochondrial function and increased oxidative stress. In inherited metabolic syndromes, reduced enzyme efficiency can lead to lactate accumulation and disturbed energy homeostasis. Background

The protein encoded by this gene belongs to the aconitase/IPM isomerase family. It is an enzyme that catalyzes the interconversion of citrate to isocitrate via cis-aconitate in the second step of the TCA cycle. This protein is encoded in the nucleus and functions in the mitochondrion. It was found to be one of the mitochondrial matrix proteins that are preferentially degraded by the serine protease 15(PRSS15), also known as Lon protease, after oxidative modification.

Rabbit Anti-ACO2 Recombinant Antibody (clone JE40-83) (CAT#: MRO-0023-CN)

We used the Rabbit Anti-ACO2 Recombinant Antibody for immunofluorescence on human fibroblast cells. The staining was sharp and localized clearly to the mitochondria, matching expected patterns. Background was minimal, and the antibody performed well under standard conditions without the need for extra optimization. Results were consistent across different batches, which gave us added confidence. Overall, a reliable choice for mitochondrial protein detection in IF applications.

Rabbit Anti-ACO2 Recombinant Antibody (clone 8H4) (CAT#: ZG-0066U)

Used the Anti-ACO2 for ELISA with recombinant human ACO2 protein. Worked reliably at 1:5000 dilution in carbonate buffer, showing good linearity from 0.1-10 ng. Tried coating with mitochondrial lysates from HEK293 cells too - needed higher antibody concentration but maintained specificity. Some background when testing liver homogenates that improved with 3% BSA blocking. Performs well for purified protein assays, though tissue samples may require extra optimization.

Recombinant Rabbit Anti-ACO2 Antibody (clone R07-7A1) (CAT#: VS3-FY17)

This recombinant rabbit anti-ACO2 antibody worked well in my WB with HEK293 and HepG2 cell lysates, showing clear bands at the expected size. Tried IHC-P on human liver tissue too—specific staining with minimal background. Didn't need much optimization for dilution. Also tested in IF on HeLa cells, gave distinct cytoplasmic localization. Solid performance for ACO2 detection in both cell and tissue samples.
