ACTA2: A Key Contributor to Vascular Fibrotic and Tumor Associated Disorders

ACTA2 encodes smooth muscle alpha-actin, an essential component of the contractile apparatus in vascular smooth muscle cells. Alterations in this gene are associated with numerous pathological disorders. Mutations can weaken blood vessel structure, leading to serious conditions such as thoracic aortic aneurysms and dissections, where arterial wall integrity is compromised. Abnormal levels of ACTA2 also contribute to hypertension by affecting vascular tone and responsiveness. In fibrotic diseases, the elevated expression of this cytoskeletal protein in myofibroblasts is linked to excessive extracellular matrix buildup, causing tissue rigidity and gradual organ failure, as observed in pulmonary fibrosis and liver cirrhosis. Additionally, increased ACTA2 expression is noted in various cancers, where it may affect the tumor microenvironment and promote invasive characteristics by altering cell adhesion and movement. The protein plays a vital role in wound healing and tissue remodeling, though its chronic activation can lead to pathological fibrosis. Its involvement in maintaining vascular contractility and cellular structure underlines its significance in both structural and signaling disruptions across cardiovascular, fibrotic, and cancerous diseases. Background

This gene encodes one of six different actin proteins. Actins are highly conserved proteins that are involved in cell motility, structure, integrity, and intercellular signaling. The encoded protein is a major constituent of the contractile apparatus and one of the two nonmuscle cytoskeletal actins that are ubiquitously expressed. Mutations in this gene cause Baraitser-Winter syndrome 1, which is characterized by intellectual disability with a distinctive facial appearance in human patients. Numerous pseudogenes of this gene have been identified throughout the human genome.

Mouse Anti-ACTA2 Recombinant Antibody (clone HHF35) (CAT#: ZG-1036F)
We used the Mouse Anti-ACTA2 Recombinant Antibody in ELISA assays with rat aortic smooth muscle cell lysates. The antibody delivered consistent and specific binding, with minimal nonspecific signal. It worked well with our existing ELISA setup and required no major adjustments. Results were reproducible across different runs, making it a dependable option for detecting smooth muscle markers in vascular-related studies.

Mouse Anti-ACTA2 Recombinant Antibody (clone 3F11-C2-G6) (CAT#: VS3-QX12)
Tested the Mouse Anti-ACTA2 in flow cytometry with primary human smooth muscle cells. It gave a clean, specific signal with minimal background, though optimal dilution needed slight tweaking. Tried it in IF with fixed fibroblasts—decent staining but required longer blocking to reduce nonspecific binding. Didn’t test WB, but for FC, it’s solid. Would use again for suspension cells but might explore other clones for tricky samples.

The mouse anti-ACTA2 recombinant antibody demonstrated robust performance in my ELISA experiments using human plasma samples, yielding distinct dose-response relationships. Tested WB on NIH/3T3 and HeLa cell lysates—sharp bands at ~42kDa without cross-reactivity. Tried IHC-P on human lung tissue too—specific staining in smooth muscle cells, minimal background. Required minimal optimization for dilution. Solid performance for ACTA2 detection in both cell and tissue-based assays.
