BCL2L1: A Guardian Against Mitochondrial Apoptosis

The BCL2 Like 1 (BCL2L1) gene, most commonly known as BCL-xL, is a highly potent member of the BCL2 family of anti-apoptotic proteins. BCL-xL is an essential protein for maintaining normal cell survival and is a primary protector against mitochondrial apoptosis. BCL-xL is a key effector of this process as it physically binds and sequesters the pro-apoptotic “effector” proteins BAX and BAK, preventing their oligomerization and pore formation in the mitochondrial outer membrane. BCL-xL also sequesters BH3-only proteins such as BAD and BIM. Many cancers are “addicted” to the high levels of BCL-xL they express, which is why BCL-xL is such an important survival factor and drug target for cancer therapy.

Background

The protein encoded by this gene is a member of the BCL-2 protein family. BCL-2 family members, which form hetero- or homodimers, are anti- or pro-apoptotic regulators that have been implicated in a broad spectrum of cellular functions. Proteins encoded by this gene are localized to the outer mitochondrial membrane, where they have been shown to regulate opening of outer mitochondrial membrane channels (VDAC). VDAC regulates mitochondrial membrane potential and thus controls production of reactive oxygen species and release of cytochrome C by mitochondria, which are potent inducers of cell apoptosis. Alternative splicing produces multiple transcript variants encoding two different isoforms. The longer isoform is an apoptotic inhibitor, and the shorter isoform is an apoptotic activator.

Mouse Anti-BCL2L1 Recombinant Antibody (clone 2H12)
I have used the anti-BCL-xL antibody from Creative Biolabs for research into BH3 mimetics and their effects on apoptosis. The WB clearly showed that these compounds could not decrease BCL-xL levels in our drug resistant cell lines which had very high expression levels.

Mouse Anti-BCL2L1 Recombinant Antibody (clone 3D1C1)
We performed IHC on tumor samples from patients using this antibody. The staining was strong and very specific and it seemed to correlate with treatment response. A good prognostic marker.
