CTNNB1: The Heart of the Wnt Signaling Pathway
Catenin Beta 1 (CTNNB1), commonly known as beta-catenin, is a protein with two very important roles. As a part of adherens junctions at the cell membrane, it binds cadherins to the actin cytoskeleton to promote cell-cell adhesion. In the cytoplasm and nucleus, it is the central effector of the canonical Wnt signaling pathway. Beta-catenin is perpetually phosphorylated by a “destruction complex” and targeted for proteosomal degradation in the absence of a Wnt signal. This destruction complex is inactivated by Wnt signaling, and allows for beta-catenin to accumulate, translocate into the nucleus, and act as a transcriptional co-activator to promote the expression of target genes involved in cell proliferation and cell fate. Activating mutations in CTNNB1 are a significant oncogenic driver in many cancers, most notably colorectal and liver cancer.

Background
CTNNB1, encoding β-catenin, is a multifunctional protein that plays a dual and critical role in cellular biology: it acts as a central component of cadherin-based adherens junctions for cell-cell adhesion and is the key effector molecule of the canonical Wnt signaling pathway, regulating gene transcription. The tight regulation of β-catenin's stability and subcellular localization, predominantly controlled by its phosphorylation status, is therefore paramount for maintaining cellular homeostasis, development, and preventing disease.
β-Catenin is a dual function protein, involved in regulation and coordination of cell-cell adhesion and gene transcription. Mutations and overexpression of β-catenin are associated with many cancers, including hepatocellular carcinoma, colorectal carcinoma, lung cancer, malignant breast tumors, ovarian and endometrial cancer.

Two separate complex-associated pools are found in the cytoplasm. The majority is present as component of an E-cadherin:catenin adhesion complex composed of at least E-cadherin/CDH1 and beta-catenin/CTNNB1, and possibly alpha-catenin/CTNNA1; the complex is located to adherens junctions. The stable association of CTNNA1 is controversial as CTNNA1 was shown not to bind to F-actin when assembled in the complex. Alternatively, the CTNNA1-containing complex may be linked to F-actin by other proteins such as LIMA1.
