DDC: The Final Step in Dopamine and Serotonin Synthesis

Dopa Decarboxylase (DDC), also known as Aromatic L-amino acid decarboxylase (AADC), is an enzyme located in the cytosol of a cell. It is responsible for the final step in the synthesis of two important monoamine neurotransmitters. DDC can catalyze both L-DOPA to dopamine and 5-hydroxytryptophan (5-HTP) to serotonin. This step is critical for proper motor function and neurotransmission. Loss of AADC due to genetic mutation causes a devastating, infantile onset movement disorder. In Parkinson's disease, inhibition of DDC is a mechanism by which L-DOPA, a dopamine precursor, can be used to maintain normal levels of dopamine.

Background

The encoded protein catalyzes the decarboxylation of L-3,4-dihydroxyphenylalanine (DOPA) to dopamine, L-5-hydroxytryptophan to serotonin and L-tryptophan to tryptamine. Defects in this gene are the cause of aromatic L-amino-acid decarboxylase deficiency (AADCD). AADCD deficiency is an inborn error in neurotransmitter metabolism that leads to combined serotonin and catecholamine deficiency. Multiple alternatively spliced transcript variants encoding different isoforms have been identified for this gene.

I used the anti-DDC antibody from Creative Biolabs in our project on Parkinson's disease. The IP staining of substantia nigra sections was specific and easily allowed visualization of dopaminergic neurons.
