Fibroblast activating protein alpha is a 170 kDa melanoma membrane bound gelatinase and is encoded by the FAP gene in humans. FAP, also known as seprase, belongs to the group SC protease and is a member of the S9B prolyl oligopeptidase subfamily. The other members of the S9B subfamily are DPPIV, DPP8 and DPP9. FAP and DPPIV are the most closely related, and they account for about 50% of the amino acids. The protein encoded by the FAP gene is a homodimer integrated membrane gelatinase belonging to the serine protease family. It is selectively expressed in reactive stromal fibroblasts of epithelial cancer, in granulation tissue of wound healing, and in malignant cells of bone and soft tissue sarcoma. This protein is thought to be involved in the control of fibroblast growth or epithelial mesenchymal interactions during development, tissue repair, and epithelial carcinogenesis. FAP is catalytically active as a 170 kD dimer and has dipeptidase and gelatinase activity. Its gelatinase activity requires glycine at the P2 position. FAP expression is seen on more than 90% of activated stromal fibroblasts in all human cancers. Interstitial fibroblasts play an important role in the development, growth and metastasis of cancer. It has been shown that targeting FAP inhibits tumor growth and growth in mice. Sibrotuzumab is a humanized monoclonal antibody used to treat cancer. It is mainly for FAP.
