GOT1: A Cytosolic Hub for Amino Acid Metabolism and Redox Balance
Glutamic-Oxaloacetic Transaminase 1 (GOT1), also known as aspartate aminotransferase 1 (AST1), is the cytosolic isoform of aspartate aminotransferase (AST). GOT1 catalyzes the reversible transamination between aspartate and α-ketoglutarate, yielding oxaloacetate and glutamate. GOT1 connects amino acid metabolism to the Krebs cycle and plays a significant role in maintaining cellular redox homeostasis. In cancer cells with KRAS mutations, GOT1 is a part of a metabolic rewiring that supports cell proliferation under nutrient-limiting conditions by maintaining redox homeostasis (producing NADPH) and enabling the synthesis of macromolecules necessary for cell proliferation. As such, GOT1 presents an attractive metabolic target for cancer therapy development.
