ICOS: A Versatile Immunotherapeutic Target

Inducible T-cell co-stimulator (ICOS, CD278), a member of the CD28 family, is upregulated upon T cell receptor (TCR) and/or CD28 co-stimulation. While its role in Th1 immunity remains debated, ICOS promotes Th2 responses. Critically, ICOS is essential for the development, survival, and follicular localization of T follicular helper (Tfh) cells, which are crucial for driving antibody affinity maturation within germinal centers. Given its influence on these key aspects of T cell function, ICOS significantly impacts the pathogenesis of numerous human diseases.

Background
