Background

Neural cell adhesion molecule 1 (NCAM1), or CD56, is a transmembrane glycoprotein in the immunoglobulin superfamily, expressed on neural cells, NK cells, T cell subsets, and some hematopoietic progenitors. It mediates cell-cell adhesion and signaling, regulating neural development and immune cell function. CD56 is overexpressed in neuroendocrine tumors (small cell lung cancer, neuroblastoma), AML, and NK/T cell lymphoma. It serves as a diagnostic marker and therapeutic target, with antibodies and CAR-T cells under development to target CD56+ tumor cells. Its expression on NK cells requires careful targeting to avoid immune-related toxicities.
