PPP3R2: The Testis-Specific Calcineurin B Isoform
Protein Phosphatase 3 Regulatory Subunit B, Beta (PPP3R2) is a highly specialized isoform of the calcineurin B regulatory subunit, which activates calcineurin by binding calcium and targeting the calcineurin catalytic subunit to membranes. The calcineurin alpha-isoform (PPP3R1) is ubiquitously expressed while PPP3R2 expression is testis- and sperm-specific. Like its somatic counterpart, PPP3R2 forms a calcium-activated complex with calcineurin's catalytic subunit, which is enriched in late stages of spermatogenesis and which has been implicated in sperm motility. Given the testis-specific expression of this protein, the calcineurin-regulated pathway containing PPP3R2 is a potential target for reversible male contraceptives with few off-target effects.

Forms a complex composed of a calmodulin-dependent catalytic subunit (also known as calcineurin A) and a regulatory Ca(2+)-binding subunit (also known as calcineurin B). There are three catalytic subunits, each encoded by a separate gene (PPP3CA, PPP3CB, and PPP3CC) and two regulatory subunits which are also encoded by separate genes (PPP3R1 and PPP3R2) (By similarity). Interacts with SPATA33 (via PQIIIT motif) (PubMed:34446558).
