SLC7A9: The Light Subunit of the Renal Cystine Transporter

Solute Carrier Family 7 Member 9 (SLC7A9), also known as rBAT, is a multi-pass transmembrane protein that acts as the light subunit of the b(0,+) amino acid transporter. It forms a disulfide-linked heterodimer with the heavy subunit SLC3A1 (rBAT) to form the functional b(0,+) amino acid transporter. The b(0,+) complex is localized to the apical membrane of proximal tubule cells, where it functions to reabsorb cystine, ornithine, lysine and arginine from the glomerular filtrate. Loss of function mutations in SLC7A9 or its associated protein SLC3A1 lead to a decreased capacity to reabsorb these amino acids, leading to very high levels in the urine, the formation of painful cystine kidney stones and chronic kidney disease. Collectively, these symptoms are known as cystinuria.

Background

This gene encodes a protein that belongs to a family of light subunits of amino acid transporters. This protein plays a role in the high-affinity and sodium-independent transport of cystine and neutral and dibasic amino acids, and appears to function in the reabsorption of cystine in the kidney tubule. Mutations in this gene cause non-type I cystinuria, a disease that leads to cystine stones in the urinary system due to impaired transport of cystine and dibasic amino acids. Alternate transcript variants, which encode the same protein, have been found for this gene. [provided by RefSeq, Jul 2011]
