CD18 (integrin beta chain-2) is an integrin beta chain protein encoded by the ITGB2 gene in humans. After binding to one of a variety of alpha chains, CD18 is capable of forming a variety of heterodimers which play an important role in cell adhesion and cell surface signaling and play an important role in immune responses. CD18 also exists as a soluble ligand binding form. Defective CD18 expression can lead to adhesion defects in circulating white blood cells in the human body and reduce the immune system's ability to withstand foreign invaders. The binding of CD18 and CD11b-d results in the formation of complement receptors (for example the macrophage-1 antigen receptor Mac-1 that binds to CD11b), which are mainly present on neutrophils, macrophages and NK cells. These complement receptors participate in the innate immune response by recognizing foreign antigen peptides and phagocytosing them, thereby destroying the antigen. In the human body, the lack of functional CD18 results in insufficient adhesion of leukocytes, a condition in which there is a lack of extravasation of leukocytes into the tissue, which means that circulating leukocytes cannot respond to foreign substances present in the tissue. This subsequently reduces the individual's immune system's ability to resist infection, making it more susceptible to foreign infection than patients infected with the functional CD18 protein. Rovelizumab is a humanized monoclonal antibody that is an experimental immunosuppressive drug. Roverizumab was developed by Icos to treat patients with hemorrhagic shock. The drug is a monoclonal antibody that suppresses the overactive white blood cells during shock.
