Protein disulfide isomerase A3 (PDIA3), also known as ERp57, is a 58kD glucose regulatory protein that is a thiol oxidoreductase chaperone belonging to the PDI family. This molecule is involved in a variety of cellular functions, including the folding of newly synthesized proteins and the assembly of major histocompatibility complex I (MHC-I). PDIA3 is expressed in human ovarian cancer, breast cancer, uterine cancer, lung cancer, gastric cancer and prostate cancer. The disulfide oxidative isomeric activity of PDIA3 is related to its own thioredoxin domain, and all members of the PDI family include at least one thioredoxin domain. PDIA3 includes four such thioredoxin regions: two active sites CXXC (named a-type region) with thioredoxin, in which cysteine has sulfhydryl and disulfide conversion activity: The other two regions lack active sites (named b-type regions).
Studies have found that PDIA3 plays an important role in regulating the proliferation of liver cancer cells and is closely related to the prognosis of liver cancer. PDIA3 regulates cell invasiveness and serves as a prognostic marker for cervical cancer. The expression of PDIA3 also affects the prognosis of gastric cancer. PDIA3 also affects the resistance of laryngeal cancer cells to radiotherapy by regulating STAT3 activity and serves as a prognostic indicator for laryngeal cancer. Consumption of PDIA3 in tumor cells will also inhibit its proliferation and increase sensitivity to ionizing radiation and chemotherapeutic drugs. Therefore, PDIA3 is expected to become a new target for anticancer treatment. In addition, the impact of PDIA3 on the biological behavior of colorectal cancer and whether it is expected to become a new tumor marker for colorectal cancer still requires us to study in the follow-up work.
