Provided is an antibody muM4TS.11 that specific for the platelet-derived growth factor receptor β. muM4TS.11 does not bind to deletion proteins containing only the first through third domains of PDGF-R β, but does bind to protein D1-4 containing the fourth domain, showing that the binding epitope is in the fourth domain. muM4TS.11 effectively inhibited the stimulation of proliferation by PDGF BB, showing the ability of these antibodies to neutralize the biological activity of PDGF BB.

CHO C4 cells expressing the human PDGFRb were preincubated with 5 m g of the indicated antibody. Addition of 100 ng of unlabeled PDGF BB served to indicate the level of nonspecific 125I-labeled PDGF BB binding. CHO C4 cells were preincubated with various concentrations of M4TS.11 (●), M4TS.15 (▲), or M4TS.22 (■) for 2 h, after which PDGF BB was added.

Cells preincubated with M4TS.11 before exposure to 125I-labeled PDGF BB and cross-linking had approximately 50% that of the level of receptor dimer as compared with cells exposed to PDGF BB in the presence M4TS.15 or no antibody.

Baboon SMC or the rabbit cornea cell line SIRC were preincubated with 10 μg/ml concentrations of the indicated antibodies for 2 h, after which PDGF BB was added.[3H]Thymidine incorporation was determined 20 h later. Proliferation of antibody-treated cells is expressed as a % of the level of proliferation observed with a nonbinding isotype-matched control antibody (Control Ab). It demonstrates that preincubation with either M4TS.11 or M4TS.22 significantly decreased PDGF BB-mediated proliferation of the baboon SMC. M4TS.11, but not M4TS.22, exhibited a similar effect on the rabbit SIRC cell line.
