ABCG1: A Key Transporter in Reverse Cholesterol Transport
ATP-binding cassette subfamily G member 1 (ABCG1) is a cholesterol transporter involved in maintaining cellular lipid homeostasis. The transporter is widely expressed in macrophages, endothelial cells, and various tissues. ABCG1's main function is to facilitate the efflux of excess cellular cholesterol, particularly to mature forms of high-density lipoprotein (HDL) particles. The transporter is thus a major player in reverse cholesterol transport, an important anti-atherogenic pathway where cholesterol is removed from the atherosclerotic plaques in artery walls by being delivered from foam cells to HDL particles. For this reason, normal ABCG1 function is essential for preventing cholesterol accumulation in the artery wall.

Background
The protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the White subfamily. It is involved in macrophage cholesterol and phospholipids transport, and may regulate cellular lipid homeostasis in other cell types. Six alternative splice variants have been identified.
