ASNS: A Key Survival Enzyme in Cancer and a Marker of Asparaginase Resistance

Asparagine Synthetase (ASNS) is a cytosolic enzyme that catalyzes the ATP-dependent conversion of aspartate and glutamine to asparagine. This is the only pathway by which human cells can produce their own asparagine. Although most normal cells can produce asparagine at levels that exceed their requirements, certain cancers, such as acute lymphoblastic leukemia (ALL), have low ASNS expression and are unable to synthesize sufficient asparagine to support cell growth. This "ASNS dependency" is the therapeutic basis for L-asparaginase therapy, which depletes serum asparagine and preferentially starves the cancer cells of this essential amino acid. However, one of the major mechanisms by which cancer cells become resistant to L-asparaginase treatment is through ASNS upregulation. ASNS is therefore an attractive biomarker of asparaginase resistance, and a target for combinatorial approaches to sensitize tumors to asparagine depletion.

Background

The protein encoded by this gene is involved in the synthesis of asparagine. This gene complements a mutation in the temperature-sensitive hamster mutant ts11, which blocks progression through the G1 phase of the cell cycle at nonpermissive temperature. Alternatively spliced transcript variants have been described for this gene.
