CAMK4: A Nuclear Decoder of Calcium Signals

Calcium/Calmodulin-Dependent Protein Kinase IV (CAMK4) is a serine/threonine kinase whose predominant localization is the nucleus. It is activated by the upstream kinase CaMKK in response to sustained calcium signaling. Once active, CAMK4's primary role is to phosphorylate a wide array of transcription factors, most prominently CREB (cAMP-response element-binding protein). This phosphorylation is a crucial step in activity-dependent gene expression in both the immune system (e.g., during T-cell differentiation) and the nervous system (e.g., during long-term memory formation). Dysregulation, particularly over-activity, of CAMK4 is linked to autoimmune conditions such as lupus.

Background

Glutamic-oxaloacetic transaminase is a pyridoxal phosphate-dependent enzyme which exists in cytoplasmic and inner-membrane mitochondrial forms, GOT1 and CAMK4, respectively. GOT plays a role in amino acid metabolism and the urea and tricarboxylic acid cycles. The two enzymes are homodimeric and show close homology. Two transcript variants encoding different isoforms have been found for this gene.
