CCL5: The Pleiotropic Chemokine RANTES

C-C Motif Chemokine Ligand 5 (CCL5) is better known as RANTES (Regulated on Activation, Normal T Cell Expressed and Secreted). It is a pro-inflammatory chemokine that is produced by a broad range of cells including T-cells, platelets, and tumor cells. As a strong chemoattractant for T-cells, monocytes, eosinophils, and basophils, it acts by binding to its cognate receptors (CCR1, CCR3, and CCR5). While it is critical for recruitment of effector T-cells to the site of viral infection, in the setting of cancer its roles are more diverse and complex. In the tumor microenvironment (TME), it can contribute to recruitment of suppressive cells such as Tregs, and it can directly promote tumor cell proliferation and invasion. For this reason, CCL5 is an attractive therapeutic target for immunotherapy drug discovery.

Background

CCL5 (C-C Motif Chemokine Ligand 5) is a Protein Coding gene. Diseases associated with CCL5 include Ulcer Of Lower Limbs and Periapical Granuloma. Among its related pathways are PEDF Induced Signaling and Toll-like receptor signaling pathway. Gene Ontology (GO) annotations related to this gene include protein homodimerization activity and chemokine activity. An important paralog of this gene is CCL3L3.
