CCL5: The Pleiotropic Chemokine RANTES
C-C Motif Chemokine Ligand 5 (CCL5) is better known as RANTES (Regulated on Activation, Normal T Cell Expressed and Secreted). It is a pro-inflammatory chemokine that is produced by a broad range of cells including T-cells, platelets, and tumor cells. As a strong chemoattractant for T-cells, monocytes, eosinophils, and basophils, it acts by binding to its cognate receptors (CCR1, CCR3, and CCR5). While it is critical for recruitment of effector T-cells to the site of viral infection, in the setting of cancer its roles are more diverse and complex. In the tumor microenvironment (TME), it can contribute to recruitment of suppressive cells such as Tregs, and it can directly promote tumor cell proliferation and invasion. For this reason, CCL5 is an attractive therapeutic target for immunotherapy drug discovery.
