CX3CR1: The Microglia-Specific Chemokine Receptor

CX3CR1, or C-X3-C motif chemokine receptor 1, is a G protein-coupled receptor (GPCR) that is strongly and selectively expressed on certain immune cell populations, including monocytes, NK cells, and most prominently, CNS-resident microglia. Its unique ligand CX3CL1 (also known as Fractalkine) is the only chemokine that is often expressed in its membrane-tethered form on the surface of neurons. The CX3CR1-CX3CL1 axis thus mediates crosstalk between neurons and microglia. Signaling through CX3CR1 keeps microglia in a quiescent, tissue surveilling state. Mutations that result in loss of CX3CR1 function lead to an increased risk of neurodegenerative disease. Thus, this pathway is critical for brain health.
