The reactivity of A5 scFv with the MRP1-derived peptide 1 as well as other MRP1-or Pgp derived peptides, strepatavidin (a) or various whole proteins (b) was tested by ELISA. MRP1 or Pgp derived peptides were immobilized onto the microtiter plates through their biotin label. Detection was with HRP-labeled anti-Myc antibody.

The binding affinity was first estimated by titration ELISA using a purified soluble A5scFv reacted with immobilized MRP1 peptide 1 (a). We also used a competitive binding assay; here we assessed the ability of the purified A5 scFv to inhibit the binding of -labeled A5 scFv to immobilized MRP1-derived peptide 1. The apparent binding affinity of the recombinant scFv was determined as the concentration of competitor (soluble purified scFv) required for 50% inhibition of the binding of the -labeled tracer (b). Error bars indicate standard deviation of triplicates of 3 independent experiments.

MRP1 expressing and MRP1 null cells (106 cells) were incubated with purified A5 scFv antibody (50 μg/ml). The reactivity was determined with a secondary FITC-conjugated anti-Myc antibody directed to the Myc tag present at the C-terminus of the A5 scFv antibody.
