a) Immunization of a mouse with p-azo-D-phenylalanine-KLH resulted in the production of stereoselective antibodies as verified in a noncompetitive ELISA using serial dilutions of serum on microtiter wells coated with p-azo-D-phenylalanine-BSA (●) and p-azo-L-phenylalanine-BSA (○), respectively. Results obtained on control-wells coated with underivatized BSA were subtracted. (b) As determined by competitive ELISA, the monoclonal antibody anti-D-AA 67.36 stereoselectively binds to the D-enantiomer of the azo-structure (●) used in varying concentrations as competitor, but not to the L-enantiomer (○). Competitive tests with D-Phe (■) and L-Phe (□) show that the antibody's affinity for the D-enantiomer of Phe is slightly lower compared to the azo-structure. Data points represent means of triplicate determinations. Missing error bars are obscured by symbols.
