At 48 hours, chLym-1 (-□-) caused a significant growth inhibitory effect over the range of 0.01–0.13 μM. Wells incubated with 0.13 μM of chLym-1 contained 90% fewer cells than untreated cells. LeLPLDB (-●-), (Leac-PLD)2PLDB (-○-), and ItPLDB (-■-) caused no growth-in-hibitory effect over the same concentration range.

  • This antibody has been reported/tested for use in Inhibition Assay.
    Inhib: Twenty (20)-, 2-, and 0.2-μM LeLPLDB, (Leac-PLD)2LPB, and ItPLDB solutions were preparedin RPMI-1640, supplemented with 10% heat-in-activated FBS. Aliquots of the 20-μM solutionswere removed and incubated with 1:100 SAHRP for 1 hour at ambient temperature before serial dilution. Raji and B35M cells were suspended at a concentration of 500,000 cells/mL in the samemedium. Negative controls were prepared, con-taining concentrations of biotin, SAHRP, and di-methylsulfoxide (DMSO) equal to that of the SHAL dilutions. One hundred (100) μL of both the SHAL solutions and the cell solutions were added in duplicate to 96-well plates. The plates were incubated at 37°C in a 5% CO₂ atmosphereand removed for counting at 24, 48, and 72 hours.At each time point, 100 μL was removed from each well and 10 μL of trypan blue was added and mixed, using gentle pipeting. Ten (10) μL of the resulting solution was added to each side of a hemocytometer, and the cells were counted using a light microscope under 100× magnification. Viable cells appeared colorless, whereasdead cells appeared blue because of uptake of try-pan blue dye. Wells containing chLym-1 at concentrations of 0.17, 0.33, 3.3, 1.7, 33, 67, and 133 nM were used as positive controls.
