The fusion protein is composed of a CD30 binding domain (HRS3-scFv) that is linked via the human IgG hinge-CH2/CH3 domain to human IL2. It was expressed in CHO and purified with affinity chromatography. The immunocytokine retains the ability to bind the CD30 as well as the biological activity of IL2. This antibody-(IL2) fusion protein retains the specificity for the CD30 antigen and the immunostimulatory function of IL2 including the activation of T and resting NK cells. HRS3scFv-Fc-(IL2) was able to induce IFN-γ secretion in SCID mice that had received subcutaneous injections of L540, a human CD30+ non-Hodgkin's lymphoma cell line, which is important for the induction of an efficient antitumor response in vivo and the shift of a T-helper type 2 (TH2) immune response in lymphoma to a TH1 response. This immunocytokine was designed for treating Hodgkin's disease (HD) anaplastic large cell lymphoma.
