Cell-binding reactivity of 4G12 MAbs on various lung carcinoma cell lines. Reactivity was determined by ELJSA as described in the Materials and Methods section. Target cells used were PC 10 squamous cell carcinoma (•), PC14 adenocarcinoma (A), PC13 large cell carcinoma (•), PC6 small cell carcinoma (A), and normal adult fibroblast (O). Absorbance of negative control as P3U1 culture supernatant and purified human IgM is <0.1.

Cell-cycle-dependent expression of 4G12 antigen on PC 10 lung squamous cell carcinoma. PC 10 cells (5 X 103 ) were stained with 4G12 MAbs [thick line) and control IgM (thin line) followed by reaction with FITC-labeled anti-human IgM and then were analyzed by FACS IV. Cells negative or positive for 4G12 expression were then separated by FACS IV with a logarithmic amplifier, cloned, and cultured for 45 days.
