Introduction

According to recent researches about coronavirus nucleoproteins (NPs or N proteins), Creative Biolabs now provides SARS-CoV-2 Nucleoprotein (NP) ELISA Kit for supporting SARS-CoV-2 detection and researches. Coronaviruses are enveloped viruses with a positive-sense RNA genome and a nucleocapsid of helical symmetry. Coronavirus NPs are required during the coronavirus RNA synthesis, and have RNA chaperone activity that may be involved in template switch. NP is the most abundant protein of coronavirus. During virion assembly, N protein can bind to viral RNA and lead to the formation of the helical nucleocapsid. Because of the conservation of N protein sequence and its strong immunogenicity, the N protein of coronavirus can be used as a detective tool.

Principles of SARS-CoV-2 Nucleoprotein (NP) ELISA Kit

The principle of this ELISA kit is based on the solid phase sandwich enzyme immunoassay technique, shown in Fig.1. This kit is to detect the novel coronavirus nucleocapsid (N) antigen in serum or plasma samples from candidates. In the beginning, a monoclonal antibody specific for SARS-CoV-2 NP has been pre-coated onto well plate strips. When samples are added to the plate, SARS-CoV-2 NP will be bound by the immobilized antibody if they are present in samples. After incubation the plate, it will be washed and a horseradish peroxidase-conjugated anti-SARS-CoV-2 NP antibody is added, producing an antibody-antigen-antibody "sandwich complex". Finally, washing unbound antibody by a TMB substrate solution and then color develops in proportion to the amount of SARS-CoV-2 NP bound and can be measured at 450 nm to observe test results.
