After growth in trypticase soy broth the bacteria were washed twice with 0.15 M NaCl. From a cell suspension with an A₅₄₆ nm of about 0.25 a 1:8 dilution was prepared. 5 μ1 of this bacterial suspension were added per well of a 24-well microscope slide, air-dried and fixed for 10 min with acetone. MAbs were applied as serial dilutions of ascitic fluid in PBS (phosphate buffered saline)ranging from 2⁵ to 2²⁴ and incubated in a humid chamber for 30 min at 37°C. After two washes with PBS samples were incubated with FITC-(fluorescein isothiocyanate)-labeled goat-anti-mouse IgG and washed under the same conditions as above. Dried probes were judged under a fluorescence microscope(Zeiss,Oberkochen).
