Mouse Anti-HIV-1 gp41 Recombinant Antibody (clone 1583) (CAT#: PABZ-068)

Recombinant Mouse Antibody (1583) is capable of binding to HIV-1 gp41, expressed in mammalian cells. The epitope can be subdivided into two minimal epitopes that correspond to residues 740-743 and residues 746-750 of gpl60. 1583 binds to the second of these (ERDRD), a sequence which is highly charged.

The S1/env/3 and HIV-1 neutralizing activity of the ascites from four hybridomas was determined. Results are expressed as the reciprocal of the mAb ascitic fluid dilution giving>90% reduction in HIV-1 titre, or neutralizing 100 TCID50 units of S1/env/3.

HIV-1-infected H9 cells either pretreated (---) or not (···) with sCD4 and uninfected H9 cells (···) were incubated fluoreehrome conjugated second antibody alone (—) or with the antibodies shown at a d dilution of either 1/120 for ascites or a concentration of 30 μg/ml for purified immunoglobulin for 2 hr at 4° with agitation, washed, and then fixed. After a further wash the cells were labeled with phycoerythrin-coupled second antibodies and then analyzed by flow cytometry on a FACScan using Consort 30 software. Each peak represents the accumulation of 10,000 gated events. Note that the staining of uninfected and HIV-infected cells with mAbs 1583 and 1908 is superimposed in this experiment.
