Figure 1 Specific inhibition of B72.3-scFv-γ receptor mediated IL-2 secretion.

Cells (10⁵ per well) of the transfected clone B72.3-scFv-γ D8.1 were cultured in BSM-coated microtiter plates in the presence of (A) B72.3 MAbb or an isotype matched IgG1 control MAb and soluble BSM or type III mucin for control, respectively. After 48 hours, supernatants were harvested and assayed for IL-2 by ELISA. Percent inhibition of IL-2 release was calculated as described in Materials and Methods. The results represent the mean of two independent experiments.

B. Clone D8.1 cells were incubated for 48 hours on plastic immobilized BSM. The cells were washed, and the cytolytic activity against Eu-labeled TAG72 + H508 tumor cells (5x10⁴ per well) and TAG720-H716 tumor cells (5x10⁴ per well) was determined in an europium release assay. The cytolytic activity against Eu-labeled TAG72 + H508 tumor cells was also determined in the presence of soluble B72.3 MAb (20 μg/mL) and of an isotype matched MAb (20 μg/mL) as control, respectively. The E:T ratio was 5:1. The columns represent the mean of triplicate measurements.
