Endothelial cells (ENDOCELL) were purchased from Kurabo Co., Ltd. (Kurashiki, Japan) as the third passage culture. The cells were grown to suhconfluency, removed from the plates by EDTA treatment, washed three times with PBS and resuspended in PBS + 2% Flow CytometryS. Normal human serum (1/100 dilution) was used as a negative control. An anti-CD59 Ab EH8 and an anti-β3 integrin Ab B6A3, both murine IgG1, were used as positive controls. For each analysis, Abs were added to 10⁶ cells at 50 μg/ml and incubated on ice for 30 min. The cells were washed three times and then incubated with FITC-conjugated anti-mouse IgG or antihuman IgG (Amersham, Tokyo, Japan) on ice for another 30 min. The cells were washed, resuspended to 10⁶ cells/ml, and analyzed by flow cytometry with EPICS-Profile (Coulter, Hialeah, FL).
