Recombinant Anti-HER1 × Anti-HER3 Bispecific Antibody (Tandem Diabody) is a bispecific fusion protein with four binding sites, two of which bind to antigens of one kind of cells and the other two to the other kind of cells. Tandem Diabodies do not carry Fc domains. So they are smaller than whole IgGs or IgG-derived BsAbs but larger than tandem scFvs. Therefore, they have a shorter serum half-life than IgGs but remain longer in the circulation compared with tandem scFvs. This BsAb is capable of blocking HER1 as well as HER3 simultaneously. BsAb-mediated simultaneous interference with two (or more) RTK signaling pathways, by inactivating either the RTKs or their ligand, can reduce the possibility of such escape mechanisms and, hence, improve therapeutic efficacy. It was designed for the treatment of head and neck cancer, colorectal cancer.
