This gene and several other Fc receptor-like gene members are clustered on the long arm of chromosome 1. The encoded protein is a single-pass type I membrane protein and contains 8 immunoglobulin-like C2-type domains. This gene is implicated in B cell development and lymphomagenesis. Stable Noncleavable Linker
Our research required a noncleavable linker to ensure payload release only occurs after lysosomal degradation. The SMCC linker in this ADC met our requirements perfectly, allowing us to accurately track the delivery of the microtubule-depolymerizing agent DM1 to the lysosomal compartment of the target cells.

  1. What is the payload and how does it work?

    A: The payload is DM1 (N2'-Deacetyl-N2'-(3-mercapto-1-oxopropyl)maytansine), a maytansinoid cytotoxin. It works by binding to tubulin at or near the vinblastine-binding site. This interferes with microtubule formation and depolymerizes existing microtubules, inducing mitotic arrest in the target cells.

  2. How does the SMCC linker release the drug?

    A: The SMCC linker is a noncleavable linker. The release of the cytotoxic drug does not occur through linker cleavage in the plasma. Instead, it is postulated to occur after the ADC is internalized via endocytosis and delivered to the lysosome, where the antibody is degraded proteolytically.
