  • Streptococcus pneumoniae Pneumolysin is a pore-forming toxin secreted by almost all strains of Streptococcus pneumoniae. Its primary mechanism of action involves binding to cholesterol in the membranes of host cells (e.g., epithelial cells, immune cells) and oligomerizing to form pores. These pores disrupt membrane integrity, leading to cell lysis (death) and tissue damage. Pneumolysin contributes to Streptococcus pneumoniae virulence in multiple ways: it damages respiratory epithelial cells, facilitating bacterial invasion into lung tissue; it lyses immune cells (e.g., neutrophils, macrophages), suppressing the host immune response; and it triggers inflammation by releasing pro-inflammatory molecules from damaged cells, which can exacerbate tissue injury (e.g., in pneumonia). The toxin is also immunogenic, and antibodies against Pneumolysin can neutralize its pore-forming activity, preventing cell lysis. Due to its role in tissue damage and immune evasion, Pneumolysin is a candidate antigen for next-generation pneumococcal vaccines-targeting it could provide protection against multiple Streptococcus pneumoniae serotypes, complementing current CPS-based vaccines.
