The Anti-PTP4A1 Antibody Prodrug, Protease Activated (VS-1025-YC109) is an antibody prodrug comprised of a recombinant mouse antibody targeting human and mouse PRL1, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to PTP4A1 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to PTP4A1 effectively. PRL1 (Phosphatase of Regenerating Liver 1), or PTP4A1, is a phosphatase that acts as an oncogene and is frequently overexpressed in various solid tumors, promoting metastasis. Its function involves promoting cell motility and invasion, making it an attractive target for therapies aimed at blocking the metastatic cascade.

- This gene encodes a member of a small class of prenylated protein tyrosine phosphatases (PTPs), which contain a PTP domain and a characteristic C-terminal prenylation motif. The encoded protein is a cell signaling molecule that plays regulatory roles in a variety of cellular processes, including cell proliferation and migration. The protein may also be involved in cancer development and metastasis. This tyrosine phosphatase is a nuclear protein, but may associate with plasma membrane by means of its prenylation motif. Pseudogenes related to this gene are located on chromosomes 1, 2, 5, 7, 11 and X.
