  • Procedure Steps
  • 1. Allow all reagents to warm to room temperature.
    2. Dilute the stock antibody to 0.1 mg/mL with reaction buffer A or B. 45 μL is sufficient to bind one 400 nm black latex vial or red latex vial.
    3. Add 40 μL of 0.1 mg/mL antibody to the 400 nm black latex vial or red latex vial and reconstitute the latex by gently pipetting up and down thoroughly. Incubate the reaction at room temperature for 15 minutes.
    4. Dilute enough 10x quencher with deionized water. For one 400 nm black latex vial, you will need exactly 1 mL, so we recommend that you prepare 1.2 mL of 1x quencher per vial, which is 120 μL 10x quencher + 1080 μL water.
    5. After 15 minutes, add 1 mL 1X quencher to stop the reaction and mix by inverting several times.
    6. Allow the reaction to quench at room temperature for 5 minutes, then transfer to a microcentrifuge tube and spin at 10,000 rpm for 9 minutes. Remove approximately 850 μL of supernatant and without resuspending the pellet, spin at 10,000 rpm for another 1 minute. Remove the remaining supernatant.
    7. Gently resuspend the pellet in 40 μL of Resuspension buffer or the storage buffer of your choice.
    8. You now have 40 μL of 1% conjugate. Dilute the conjugate as needed for your application.

  • Product Precautions
  • We recommend that the antibody to be labeled should be in a solution of 10-50 mM MES, HEPES, or MOPS at pH 6-7 (without any additional components such as salt) prior to conjugation.

  • Kit Storage
  • Store kit at -20°C immediately upon receipt.
  • Conjugate Storage
  • It is recommended to store any conjugate at 4°C. The resuspension buffer added at the end of the conjugation reaction is a good storage buffer. Do not store conjugates at -20°C. The determining factor for conjugate stability is the antibody itself, as it will degrade first. Therefore, if your antibody is stable, the conjugate will also be stable.
