Background

This gene encodes one of seven subunits of the human Arp2/3 protein complex. The Arp2/3 protein complex has been implicated in the control of actin polymerization in cells and has been conserved through evolution. The exact role of the protein encoded by this gene, the p34 subunit, has yet to be determined. Two alternatively spliced variants have been characterized to date. Additional alternatively spliced variants have been described but their full-length nature has not been determined.

Component of the Arp2/3 complex composed of ACTR2/ARP2, ACTR3/ARP3, ARPC1B/p41-ARC, ARPC2/p34-ARC, ARPC3/p21-ARC, ARPC4/p20-ARC and ARPC5/p16-ARC (PubMed:9230079, PubMed:11741539). Interacts with SHANK3; the interaction probably mediates the association of SHANK3 with the Arp2/3 complex (By similarity). Interacts with DNAI3; this interaction reduces binding of the Arp2/3 complex to the VCA domain of nucleation promoting factors (PubMed:32128961).

Mouse Anti-ARPC2 Recombinant Antibody (clone CBLS-148) (CAT#: MOB-0855MZ)
We've been using CBLS-148 for about 8 months in our cytoskeleton studies. Works like a charm in WB with A549 cell lysates at 1:800 dilution - gives that crisp 34kDa band every time. Took some tweaking to get clean IF results in our patient-derived fibroblasts. Surprisingly good in cross-linking IPs too, though we noticed slight degradation if left at room temperature too long. Not the cheapest option, but the consistency across batches has saved us so much troubleshooting time that it's worth every penny.

Recombinant Rabbit Anti-ARPC2 Antibody (clone R04-1B6) (CAT#: VS3-FY98)
The R04-1B6 clone has worked well for IF in our lab. We got clean cytoplasmic staining in fixed A549 cells at 1:300 dilution, though it required 0.1% Triton permeabilization for 20 minutes. The signal was weaker in primary neurons compared to cell lines. Batch consistency has been good over six months of use. A reliable mid-range option for basic ARPC2 localization studies.
