ASMT: A Vital Regulator in Sleep Mood and Neurodegenerative Disorders

ASMT is a key enzyme involved in the final step of melatonin biosynthesis, and its dysfunction has been increasingly associated with a range of neurological and psychiatric conditions. Altered enzymatic activity can disrupt circadian rhythm regulation, which plays a critical role in sleep disorders, particularly insomnia and delayed sleep phase syndrome. Impaired melatonin production has also been linked to mood disorders such as depression and bipolar disorder, where disruptions in the sleep-wake cycle and hormonal balance contribute to symptom severity. In autism spectrum disorders, genetic variants affecting this enzyme’s function are believed to contribute to abnormal melatonin levels, which may influence behavioral symptoms and sleep disturbances commonly observed in affected individuals. Additionally, the enzyme’s role in oxidative stress regulation and neuroprotection suggests its relevance in neurodegenerative diseases, including Alzheimer’s and Parkinson’s, where melatonin has been studied for its antioxidant and mitochondrial-stabilizing properties. Background

This gene belongs to the methyltransferase superfamily, and is located in the pseudoautosomal region (PAR) at the end of the short arms of the X and Y chromosomes. The encoded enzyme catalyzes the final reaction in the synthesis of melatonin, and is abundant in the pineal gland. Alternatively spliced transcript variants have been noted for this gene.

We used the Rabbit Anti-ASMT Recombinant Antibody for Western blot analysis on human pineal gland lysates. The antibody showed clear, specific bands with very low background, which made interpretation straightforward. It integrated well with our existing protocol and didn’t require much optimization. Signal intensity was strong and reproducible across different experiments. Overall, a dependable reagent that helped us validate ASMT expression in our tissue samples without any technical issues.

Rabbit Anti-ASMT Recombinant Antibody (clone DS266AB) (CAT#: MOR-0266)
This rabbit anti-ASMT antibody worked great in my WB experiments with HEK293 and HeLa cell lysates. Showed sharp bands without cross-reactivity. Tried IHC-P on mouse liver tissue too—excellent specificity, minimal background. Required no optimization for dilution. Definitely recommend for ASMT detection in both cell and tissue samples.
