ATOX1: The Copper Chaperone for the Secretory Pathway

Antioxidant Protein 1 (ATOX1) is a small cytosolic copper chaperone that is responsible for binding copper ions from the C-terminus of the Ctr1 copper importer at the plasma membrane and shuttling the copper through the cytosol to the trans-Golgi Network (TGN), where it is released to the copper-transporting ATPases ATP7A and ATP7B for loading into copper-dependent enzymes (i.e., those involved in collagen cross-linking, pigmentation, etc.). ATOX1 also has a nuclear localization signal and can act as a transcription factor, and its overexpression in cancer has been associated with increased proliferation and angiogenesis.

Background

This gene encodes a copper chaperone that plays a role in copper homeostasis by binding and transporting cytosolic copper to ATPase proteins in the trans-Golgi network for later incorporation to the ceruloplasmin. This protein also functions as an antioxidant against superoxide and hydrogen peroxide, and therefore, may play a significant role in cancer carcinogenesis. Because of its cytogenetic location, this gene represents a candidate gene for 5q-syndrome.
