BAG1: A Co-Chaperone Linking Chaperones to the Proteasome

Cellular chaperones, like Hsp70, are key components of the protein quality control system that maintain cellular proteostasis and homeostasis. BAG1 is a co-chaperone that directly interacts with and regulates Hsp70 activity. Its main function is binding the ATPase domain of the Hsc70/Hsp70 chaperone and serving as a nucleotide exchange factor. In this way, BAG1 controls chaperone activity and its ability to fold client proteins. It also contains a ubiquitin-like domain, allowing it to link the chaperone complex directly to the proteasome and mediate the degradation of misfolded proteins. BAG1 also interacts with and potentiates the anti-apoptotic function of BCL2 and can modulate the activity of steroid hormone receptors.

Background

The BCL2 oncogene is a membrane protein that inhibits a step in a pathway leading to apoptosis (programmed cell death). The protein encoded by this gene binds to BCL2 and is therefore known as BCL2-associated athanogene. This gene product augments BCL2 anti-apoptotic function, and appears to function as a bridge between growth factor receptors and anti-apoptotic machinery. This mRNA encodes multiple protein isoforms by utilization of a non-AUG (CUG) initiation codon, and three alternative downstream AUG initiation codons. A related pseudogene has been identified on the X chromosome.
