BCL2L2: An Anti-Apoptotic Protein with Roles in Hematopoiesis and Cancer
BCL2 Like 2 (BCL2L2), also known as BCL-w, is a pro-survival member of the BCL2 family of proteins. It is expressed on the outer mitochondrial membrane, similar to its close homologs BCL2 and BCL-xL. BCL-w exerts its anti-apoptotic effects by binding and sequestering pro-apoptotic BCL2 family members, including BAX, BAK, and BH3-only proteins. BCL-w is critical for hematopoietic cell survival and is required for normal spermatogenesis. It is also overexpressed in certain cancers, including gastric and colon cancer, where it promotes tumor cell survival and resistance to chemotherapy.

Background
The protein encoded by this gene is a member of the BCL-2 family of proteins. Hetero- or homodimers of these proteins function as regulators of apoptosis, with some members acting in anti-apoptotic and others in pro-apoptotic ways. Expression of this gene in cells has been shown to result in decreased cell apoptosis in cytotoxic conditions. Analysis of the related gene in the mouse suggests a role in the survival of NGF- and BDNF-dependent neurons. Mutation and knockout studies of the mouse gene showed an essential role in adult spermatogenesis. Alternative splicing results in the production of multiple transcript variants. Read-through transcription from this gene into the neighboring downstream PABPN1 (poly(A) binding protein, nuclear 1) gene also occurs.
