BDKRB1: An Inducible Receptor for Chronic Pain and Inflammation

Bradykinin Receptor B1 (BDKRB1), a G protein-coupled receptor (GPCR), is inducible and not constitutively expressed in healthy tissues but is rapidly upregulated in response to inflammatory mediators such as cytokines and bacterial endotoxins. The receptor is activated by des-Arg-bradykinin, a metabolite of the pro-inflammatory mediator bradykinin. The resulting pro-inflammatory activities that are mediated by BDKRB1 include vasodilation, increased vascular permeability, and, most importantly for pain research, the sensitization of sensory neurons to cause chronic pain and hyperalgesia. As an inducible target, BDKRB1 represents a very specific mechanism for therapeutic intervention during chronic inflammatory and pain states with likely reduced side effects compared to antagonists for targets that are expressed constitutively.
