C1QB: An Essential Subunit of the C1q Complex

Complement C1q is a heterotrimeric glycoprotein, comprised of six identical copies of the A (C1QA), B (C1QB), and C (1QC) chains. The C1QB chain is a critical subunit that co-assembles with the A and C chains to form the “tulip head” structure that can recognize and bind to antibody Fc regions. Deficiencies or mutations in C1QB can preclude assembly of a functional C1q complex, which leads to defects in immune complex clearance and autoimmune diseases, most notably systemic lupus erythematosus (SLE).

Background

C1q associates with the proenzymes C1r and C1s to yield C1, the first component of the serum complement system. The collagen-like regions of C1q interact with the Ca(2+)-dependent C1r(2)C1s(2) proenzyme complex, and efficient activation of C1 takes place on interaction of the globular heads of C1q with the Fc regions of IgG or IgM antibody present in immune complexes.
