C1QC: A Core Component of the C1q Sensor

Complement C1QC is one of three polypeptide chains (C1QA, C1QB, and C1QC) that form obligate trimeric complexes in the production of the C1q complex. Six copies of the C1QC chain assemble with six copies each of C1QA and C1QB chains to form the mature C1q molecule. C1q is the sensor of the classical complement pathway, and each chain is critical for structure and binding activity of the molecule. In fact, deletion of any of the three chains, including C1QC, is a high-risk factor for the development of severe systemic lupus erythematosus (SLE) due to the inability to clear apoptotic debris in the absence of the C1q-mediated pathway.

Background

C1q associates with the proenzymes C1r and C1s to yield C1, the first component of the serum complement system. The collagen-like regions of C1q interact with the Ca(2+)-dependent C1r(2)C1s(2) proenzyme complex, and efficient activation of C1 takes place on interaction of the globular heads of C1q with the Fc regions of IgG or IgM antibody present in immune complexes.
