CA14: A Transmembrane pH Regulator in the Brain
Carbonic Anhydrase 14 (CA14) is a member of the alpha-carbonic anhydrase superfamily. It is unique among the well-studied cytosolic isoforms in that it is a Type I transmembrane protein, with the catalytic domain facing the extracellular side of the cell. CA14 is also highly expressed in brain, spinal cord, and retina. As a result, by catalyzing the hydration of CO2 to bicarbonate and protons in the extracellular space, CA14 is predicted to be a key regulator of local pH in the synaptic cleft and other extracellular compartments. In turn, extracellular pH will influence the activity of a large array of pH-sensitive ion channels and receptors and as such, CA14 may play an important role in neuronal excitability.

Background
Carbonic anhydrases (CAs) are a superfamily of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide and are involved in many biological processes, such as respiration, calcification, acid-base regulation, bone resorption, aqueous humor, cerebrospinal fluid, saliva and gastric acid production. They are widely distributed in tissues and organs, and show large diversity in their subcellular localization. CA XIV is predicted to be a type I membrane protein. The CA isoform that is most similar to CA XIV based on sequence similarity is the other transmembrane CA isoform, CA XII, and they have been shown to have different patterns of tissue-specific expression suggesting that they may have distinct physiologic roles.
