CA3: The Muscle and Adipose Carbonic Anhydrase

Carbonic Anhydrase 3 (CA3) is a cytosolic isoform of the alpha-carbonic anhydrase family. CA3 is a relatively inefficient catalyst for the hydration of CO2 in comparison to some other CA isoforms. However, it is one of the most abundant proteins in slow-twitch (type I) skeletal muscle fibers and adipocytes (fat cells). The most notable feature of CA3 is that it is much less sensitive to inhibition by sulfonamides, which are the classic CA inhibitors. The biological role of CA3 is likely to be protection against oxidative stress, given that it has a high reactivity with reactive oxygen species and aids in their detoxification. This protects muscle and fat cells from oxidative damage that can occur during high metabolic activity.

Background

Carbonic anhydrase III (CAIII) is part of a multigene family (there are at least six separate genes) that encode carbonic anhydrase isozymes. Carbonic anhydrases are a class of metalloenzymes that catalyze the reversible hydration of carbon dioxide, and are expressed differentially across a number of cell types. Expression of the CA3 gene is strictly tissue specific, and found at high levels in skeletal muscle and much lower levels in cardiac and smooth muscle. A subset of Duchenne muscle dystrophy carriers have an elevated CA3 level.
