CALM3: A Calmodulin Gene Linked to Cardiac Arrhythmias
Calmodulin 3 (CALM3) is a gene that encodes the calmodulin protein. Calmodulin is a highly conserved, small intracellular protein that functions as a calcium sensor and is encoded by three identical genes: CALM1, CALM2 and CALM3. While these genes are identical in the protein sequence they encode, they are transcribed from distinct genetic loci and are subject to independent transcriptional regulation as they are found on separate chromosomes. Calmodulin is an essential and highly conserved protein necessary for life. It interprets intracellular calcium signals and translates them into downstream effects by activating an enormous number of enzymes and ion channels in the cell. Mutations in the CALM3 gene alone have been found to cause severe cardiac diseases, including long QT syndrome and catecholaminergic polymorphic ventricular tachycardia (CPVT), suggesting a non-redundant role for each of the three calmodulin genes.

Background
The gene encodes a member of a family of calcium-binding proteins and acts as an enzymatic co-factor. The activity of this protein is important for cell cycle regulation and cytokinesis. Multiple alternatively spliced transcript variants have been observed for this gene.
