CAMKK2: A Master Kinase Linking Calcium and Energy Metabolism
Calcium/Calmodulin Dependent Protein Kinase Kinase 2 (CAMKK2) is a highly promiscuous serine/threonine kinase that serves as a major hub in cellular signaling pathways. It is activated in response to increases in intracellular calcium levels in a calmodulin-dependent manner. The activated CAMKK2 has two major downstream targets: (1) phosphorylation and activation of CaMKI/IV pathway, and (2) CAMKK2 is a major upstream kinase for AMP-activated protein kinase (AMPK), the master regulator of intracellular energy homeostasis. In this way, CAMKK2 links calcium signaling to AMPK directly and functions as an important mediator of metabolism, inflammation, and cell growth. It is overexpressed and tumor-promoting in a number of cancers, including prostate cancer, and is a major target for therapeutic intervention.
