CARTPT: The CART Neuropeptide Precursor

The CART Prepropeptide (CARTPT) encodes several peptides that are proteolytically processed and then collectively referred to as Cocaine- and Amphetamine-Regulated Transcript (CART) peptides. CART peptides are highly expressed in the hypothalamus, the brain’s “feeding center,” where they act as a powerful anorexigenic (appetite suppressing and energy expenditure stimulating) signal. They are also present in other brain regions that mediate rewarding and psychosocial actions of psychostimulants. This peptide system is a key target for novel anti-obesity and anti-addiction therapeutics.

Background

This gene encodes a preproprotein that is proteolytically processed to generate multiple biologically active peptides. These peptides play a role in appetite, energy balance, maintenance of body weight, reward and addiction, and the stress response. Expression of a similar gene transcript in rodents is upregulated following administration of cocaine and amphetamine. Mutations in this gene are associated with susceptibility to obesity in humans.
